I found this book once called The Hypochondriac's Guide to Life. And Death, by Gene Wiengarten. It wasn't really fiction, but to me, at least, it was pretty funny. It also has some very good theories.
We're whalers on the moon
We carry a harpoon
But there ain't no whales so we tell tall tales
And sing a whaling tune

I generally find Dave Barry to be very funny, though I believe most of his works are essays.

Another good humorous Non-fiction writer is P.J. O'Rourke, Parliment of Whores and Eat the Rich are two of my favorites by him.

I know you said no Pratchett, but I just dived into Discworld. I've read up to Men at Arms and moved onto Soul Music now. Very enjoyable all that nonsense!

Apathy and Other Small Victories by Paul Neilan. I read it when I was out sick once and ended up hurtiung myself laughing. Nice quick read and very humorous.

The Twilight Series. Not supposed to be funny but they are hilarious. Especially since you can then effectively ridicule all the stupid Twi-tards to death.

"Love and other near death experiences" i think by mil milington - very funny book if you like douglas adams but dislike his meandering style.
